猴痘病毒
猴痘病毒(Mpox virus,MPXV)與天花病毒一樣,屬于正痘病毒屬。其形態(tài)與正痘病毒一致, 外形為圓角磚形或卵圓形,大小為200~400nm。磚形或卵圓狀,外被脂蛋白膜或封套,中間具有兩個含蛋白質的側體,有1個厚膜核心,核心內含很大的雙鏈DNA基因體。M痘病毒的基因組為雙鏈DNA,長約197kb,基因組末端包含一個相同但方向相反的末端反向重復序列。該病毒包含190個開放閱讀框,其中4個位于末端反向重復序列中。猴痘病毒核心蛋白為A29L, A35R, B6R, M1R, H3L

真核表達猴痘核心蛋白及相關中和抗體Monkeypox virus/MPXV A29L
A29L是細胞內成熟病毒(IMV)表面包膜蛋白(與牛痘病毒VACV,A27同源)。溶瘤痘苗病毒(VV)屬于痘病毒科中一擁有192kb雙螺旋DNA遺傳物質的病毒,會在細胞質中繁殖出具有鞘膜的病毒,進而感染其他不同種的哺乳動物細胞及個體.VV中的A27L基因能表達出一14kDa大小的鞘膜蛋白,此蛋白能與細胞表面上的硫酸乙酰肝素(Heparan Sulfate)作用使病毒跟細胞結合并且可催化細胞融合。

Monkeypox virus/MPXV A35R

Monkeypox virus/MPXV B6R

Monkeypox virus/MPXV E8L
E8L是病毒入侵宿主細胞重要基因,在病毒入侵宿主完成復制的過程中作為MV的細胞表面結合蛋白與細胞表面的硫酸軟骨素結合,以提供病毒粒子附著在靶細胞上。有研究表明利用siRNA技術能有效拮抗Monkeypox virus的A6R和E8L基因,由此構建siA6-a和si8 -d能破壞MPV復制。
